gRNA Tool

"gRNA Tool" searches for guide RNA sequences for S. pyogenes (20mer + NGG PAM) in the given target (fasta or GenBank format). It outputs unique 23mer, corresponding target sequence (reverse complement if "-" orientation), GC content (not including PAM), number of off-targets for 1 bp and 2 bp mismatches, and the sequences of off-targets. The output is sorted by the cutting position of the target.

The graphic view displays sense-strand gRNA sequences at the top and reverse-complement (RC) sequences at the bottom.

• If there are more than 20 candidates, the tool will exclude gRNA sequences that have (1) two or more 1 bp mismatch off-targets or (2) five or more off-targets with 2 bp mismatches.
• If there are more than 50 candidates, the tool will exclude gRNA sequences that have (1) any 1 bp mismatch off-targets or (2) three or more off-targets with 2 bp mismatches.
